This study aimed to assess the pharmacokinetics of sildenafil citrate (Viagra) in healthy men and in patients with pulmonary arterial hypertension (PAH).
A single-blind, randomized, controlled, dose-escalation, single-dose, single-center, double-blinded, double-masked trial with 100 participants was conducted in 30 men and women with a mean age of 52 years.
The pharmacokinetic parameters of sildenafil were found to be different in men and women with PAH: there was no statistically significant difference in serum concentrations of sildenafil, total sildenafil, and sildenafil analogs (50% and 75% bioequivalence) in healthy volunteers and in patients with pulmonary arterial hypertension (PAH), when compared to the results of sildenafil plasma concentration (C) and total plasma concentration (T), in men with PAH, when compared to healthy volunteers.
The pharmacokinetics of sildenafil in healthy volunteers was different, with an area under the plasma concentration versus time curve of 25% and 30%, respectively, for sildenafil, and a value of 44% for sildenafil analogs (50% bioequivalence). The pharmacokinetics of sildenafil in patients with PAH was also different.
